Mutations in mTOR pathway genes lead to malformations of cortical development that are among the most common causes of epilepsies requiring resective surgery. The mTOR Variant Validation Project (MVVP; Peter Crino, PI) proposes to systematically investigate the pathophysiological mechanisms. The MVVP variant database documents >250 potentially pathogenic variants in mTOR pathway genes identified in epilepsy patients, which are the basis of our investigation.
